Actin-myosin tears it up
نویسنده
چکیده
Actin-myosin tears it up aspase cleavage of nuclear lamin proteins was thought to be sufficient to cause disintegration of the nucleus during apoptosis. Now, Croft et al. (page 245) show that the actin-myosin cytoskeleton generates a pulling force required to tear apart the structure. When studying the role of actin and myosin in plasma membrane blebbing and cell contraction during apoptosis, the team noticed that if they inhibited ROCK I, a kinase that increases actin contractility and is activated by caspase cleavage, DNA was efficiently fragmented but did not end up in the expected biochemical fractions. The team hypothesized that ROCK I and the actin cytoskeleton might be involved in apoptotic nuclear breakdown. In the current study, Croft et al. found that after blocking ROCK I activity and thus myosin light chain (MLC) phosphorylation, the nuclear envelope remained intact. Thus, the actin-myosin cytoskeleton is required for apopC

Modulation of gelsolin-induced actin-filament severing by caldesmon and tropomyosin and the effect of these proteins on the actin activation of myosin Mg(2+)-ATPase activity.
We have investigated the cumulative effects of three smooth-muscle actin-binding proteins, gelsolin, caldesmon and tropomyosin, on actin activation of myosin Mg(2+)-ATPase activity under low-ionic-strength conditions. A combination of tropomyosin (at a stoicheiometric ratio to actin) and gelsolin (at a molar ratio to actin of up to 1:100) showed essentially additive stimulatory effects that wer...
متن کاملConformationally trapping the actin-binding cleft of myosin with a bifunctional spin label.
We have trapped the catalytic domain of Dictyostelium (Dicty) myosin II in a weak actin-binding conformation by chemically crosslinking two engineered cysteines across the actin-binding cleft, using a bifunctional spin label (BSL). By connecting the lower and upper 50 kDa domains of myosin, the crosslink restricts the conformation of the actin-binding cleft. Crosslinking has no effect on the ba...
متن کاملInteraction of actin and myosin in the abscence and presence of ATP.
The binding between F-actin and myosin has been studied by analyzing the amount of radioactively labelled (by means of 14C-NEM) F-actin and myosin in the formed unsoluble actomyosin pellet after centrifugation of the reaction mixture. In the absence of ATP, the amount of actin bound to myosin varies, depending on the amounts of actin and myosin present, between 0.18 mg actin/mg myosin (2 actin ...
متن کاملThe working stroke upon myosin-nucleotide complexes binding to actin.
For many years, it has been known that myosin binds to actin tightly, but it had not been possible to devise a muscle fiber experiment to determine whether this binding energy is directly coupled to the working stroke of the actomyosin crossbridge cycle. Addressing the question at the single-molecule level with optical tweezers allows the problem to be resolved. We have compared the working str...
متن کاملThe stepping pattern of myosin X is adapted for processive motility on bundled actin.
Myosin X is a molecular motor that is adapted to select bundled actin filaments over single actin filaments for processive motility. Its unique form of motility suggests that myosin X's stepping mechanism takes advantage of the arrangement of actin filaments and the additional target binding sites found within a bundle. Here we use fluorescence imaging with one-nanometer accuracy to show that m...
